• o Chromatography Systems
The chromatographic systems are calibrated using either the
external of the internal standard techniques . Specific methods
may impose variations and/or different acceptance criteria upon
these two techniques_ These specific requirements are found in
the CHEMUEST SOP covering the particular method in question.
1
a External Standard Calibration Procedure=
For each analyte, or group of analytes, flue concentration
levels of standards are prepared by adding aliquots of one or
more stock standards to ualumetric flasks and diluting to
1 volume with an appropriate solvent. One of the standards
should be at a concentration near the method detection limit.
The other concentrations should define the working range of
the system .
Each of the calibration standards is introduced into the
chromatography system using the technique to be used for
introduction of actual samples Cie_ 2- to S-ul liquid
Injections, purge IL trap, etc _) . Both a Calibration Curue,
and a series of Calibration Factors (Cr,$) at each standard
concentration, is calculated for each analyte .
o The Calibration Curve is a plat of amount introduced
us . detector response .
amount introduced
o The CF - ------------------w------------ (for nultiresponse
' total response
compounds use the total area of all peaks used for
quantitation) .
o If the percent relative standard deviation CXRSO)
between the cr,s is less than 2OX over the working
r range, linearity through the origin can be assumed and
an average CF can be used for quantitation .
o If the above criteria is not net, the Calibration
Curue can be used for quantitation if the residual,
r, is greater than O .99S .
o If neither criteria is net, the standard analyses
must be repeated if quantitation is to be performed .
o If the quantitation criteria are not met,
documentation of the ability to sec the required
r minimum detectable concentration is sufficient to
determine the presence or absence of target compounds.
18
