All sample collection bottles are sterilized with an <br /> autoclave. Autoclave strips are required for every batch and <br /> must indicate that the sterilization process is complete. <br /> Sample bottles are dated with the sterilization date and <br /> stored appropriately. All broth tubes, confirmation sticks, <br /> media, and dilution water are sterilized in the same manner <br /> - and stored at room temperature or in the refrigerator <br /> according to the methodology. Glassware for filtering is <br /> sterilized by ultraviolet light exposure. Tongs used to <br /> transfer the filters are flame sterilized. <br /> Incubator, waterbath, and refrigerator temperatures are set <br /> according to methodology and daily records are documented for <br /> -- all. The thermometer's accuracy is checked yearly against a <br /> National Bureau of Standards registered thermometer. <br /> Sterile dilution water is plated and incubated weekly to <br /> check for contamination. The results are kept in the <br /> analysts' notebook. Laboratory deionized water is tested <br /> yearly for water suitability according to EPA methodology and <br /> monthly for a heterotrophic plate count. The results from <br /> these cross checks are documented in the analysts' notebook. <br /> ~ Bioassays are performed in accordance with California <br /> Department of Fish and Game Water Pollution Control <br /> Laboratory's "Static Acute Bioassay Procedures for Hazardous <br /> Waste Samples" 16 September, 1987 for hazardous waste samples <br /> 1 and in accordance with California Department of Fish and Game <br /> State Water Resources Control Board's "Guidelines for <br /> Performing Static Acute Toxicity Fish Bioassays in Municipal <br /> and Industrial Wastewaters" July 1976 for effluent <br /> monitoring. <br /> Bioassays are performed in a temperature controlled room <br /> reserved only for bioassays. The temperature is monitored by <br /> a continuous temperature recorder. The charts are kept in <br /> the temperature log-book. <br /> Synthetic freshwater dilution water is made from deionized <br /> water and reagent grade chemicals according to the <br /> methodology. Soft water and very hard water are stored in <br /> 200L tanks and checked before use to ensure that the <br /> alkalinity and hardness measurements fall within the <br /> specified ranges appropriate for the test species.
