Sep-21 -9$ 05 : iup
NUTRIENT ASSAYS and pH RESULTS
PROTOCOL: Colorimetric assays were performed for the determinations of ammonia,
nitrate and ortho-phosphate levels in water samples, per EPA manual
colorimetric protocols using Hach reagents and a Gilford 340 VIS/[JV
digital spectrophotometer The pH was measured with a Coming digital
pH meter Mean pH values were obtained from triplicate measurements
All assays conform to California CLP and Standard Water g Wastewater
analytical method specifications.
Ammonia Nitrate Phosphate
SAMPLE SAMPLE (mg/L) (mg/L) (mg/L)
GW-1 12/14/95 030 020 065 68
GW-2 12/14/95 350 3 5 700 68
BR-2 12/14/95 210 190 500 68
COMMENTS: The enumeration data indicate that hydrocarbon degraders were aresent in the
groundwater prior to inoculation, but the levels were moderate The limiting
factors were probably ammonia nitrogen and oxygen The inoe alation of the
batch culture increased ambient levels of hydrocarbon-degrading bacteria nearly
a hundred fold to over a million colony forming units per milliter Ammonia
nitrogen and ortho-phosphate levels were also increased approximately 10 fold,
as intended It should be noted that phosphate is readily adsorbec onto the soil
micelles as well as reacting with bivalent cations in the water To provide
adequate amounts of available phosphorous, it is important to amend the ground
water with an excess of soluble ortho-phosphate The batch inoculation also
increased the nitrate nitrogen levels slightly, but the concentration; of nitrate in
the groundwater did not exceed the drinking water standard of 10 mglL
Generally, proper control of ammonia nitrogen amendments to the ground water
will prevent the accidental increase of nitrate levels to greater thai the 10 ppm
drinking water limit The inoculation did not alter the pH of the groundwater,
which remains near optimal for in situ bioremediation of petroleum
Assays were performed by Sue Arve, M S , Director of Technical Services
JRAL t
oo9g .
i '4~ Randall von Wedel, PhD
Sue Arve, M S
Director of Technical Services
