Certification is usually accomplished utilizing instrumentation, chemical indicators, and
biological indicator to verify that appropriate physical parameters are satisfactory and
that resistant microorganisms are destroyed. It is highly recommended that biological
indicators be used rather than those chemical temperature.
3. Chemical indicators are of limited value in verifying the decontamination of
biohazardous waste. Chemical indicator inks printed on waste packaging materials
inteneded for autoclaving provides a color change that serves only to distinguish treated
waste from waste requiring treatment, i.e., failure of an indicator to change to its signal
color after the process demonstrates immediately that the equipment has
malfunctioned. Temperature indicator strips that change color indicates only that the
outside of the container was exposed to a certain temperature, they do not indicate
internal temperature or exposure times and therefore, do not indicate that the waste has
been sterilized. They do indicate, however, processing time rather than treatment
effectiveness.
4. A recording thermometer must be used to ensure that a sufficiently high temperature is
maintained for an adequate time during the cycle. Failure to attain or maintain operating
temperature may indicate mechanical failure. .
5. It is highly recommended that each load contain a biological indicator (Bacillus
sterarotheromphilus) to determine that the autoclaved biological wastes have been
sterilized at proper temperature, time and pressure.
1. Biological commodities requiring registration must be autoclaved in the primary
laboratory unless otherwise authorized..
2. All commodities being autoclaved must be accompanied on a quarterly basis by a
biological and each cycle by a time/thermal or color indicator and must be placed in the
autoclave by a member of the responsible group will see that temperature and pressure
is up to desired levels and the unequivocal instructions for sterilization are posted
before leaving the area.
3. Contaminated dead animals, animal wastes or tissues wastes must be placed into
sealed, properly labeled, doubled plastic bags, and put into a lidded leak proof, metal
container prior to autoclaving.
1. All commodities must be delivered in a lidded, leak proof, secondary container and
each secondary container must be so labeled as to indicate the laboratory of origin, the
most hazardous microorganism present and that the material has not been autoclaved.
2. If biological commodities requiring registration are involved, a member of the
responsible research group delivering the material to be autoclaved must place the
secondary container in the autoclave.
file://C:\Documents and Settings\Owner\My Documents\P&PUnfection Control 2005\Bioha... 5/5/2005
